pYMB essentials+[13-R]+[Y-CHAMP]

pYMB indicates pUC19 with ori+Pmsp1+rep. In this construct, 13-R and Y-CHAMP are ligated in the downstream of ori+Pmsp1+rep, aimed at serving as optogenetic wireless neurostimulator with OFF state (no BRET between EYFP and rLuc) as default mode. This design is made in case EYFP fusion on N-terminal of mms13 has negative effects on its folding. But according to the opposite location of EYFP on CHAMP, this part is intended for a negative control, also testing the orientation of CHAMP on the effect of BRET between EYFP and rLuc.

Fig. 8: In this last construct, only r-luciferase anchored on Mms13’s C-terminus. It will light up in yellow light (with 580 nm wavelength) due to the luciferase reaction, instead of the green light induced from BRET phenomenon (540 nm wavelength).
